Schlenk, F. (Argonne National Laboratory, Argonne, Ill.), and J. L. Dainko. Action of ribonuclease preparations on viable yeast cells and spheroplasts. J. Bacteriol. 89:428-436. 1965.-Recrystallized pancreatic ribonuclease releases more than half of the ultraviolet-absorbing cellular constituents of Candida utilis and Saccharomyces cerevisiae. The presence of salts and buffer interferes with the action of the enzyme on the cytoplasmic membrane, and intracellular constituents show a similar inhibitory effect. Polymerized ribonuclease and conformational isomers cannot penetrate the pores of the yeast cell wall, but they are effective on the membrane of spheroplasts. All forms of the enzyme interfere strongly with the viability of the cells.

The Extractability of Inner-Membrane Proteins from Salmonella typhimurium Intact Cells, Spheroplasts and Inner-Membrane Fragments by Non-Denaturing Detergents
The effect of Triton X-100, Na cholate and Tween 80 on the solubilization of integral membrane proteins in intact cells, spheroplasts and inner-membrane fragments of Salmonella typhimurium was studied. The detergents were used in various concentrations (1.6 to 64 mM) and cytochromes b and d were used as marker to monitor the solubilization of membrane-bound proteins. Results showed that no inne...

Untreated cells exhibited well-defined cell walls, w...
متن کاملMacromolecule synthesis in yeast spheroplasts.
Conditions have been established for the preparation of spheroplasts of Saccharomyces cerevisiae which are able to increase their net content of protein, ribonucleic acid (RNA), and deoxyribonucleic acid (DNA), several-fold upon incubation in a medium stabilized with 1 m sorbitol. The rate of RNA and protein synthesis in the spheroplasts is nearly the same as that occurring in whole cells incub...
متن کاملReconstitution of colicin E2-induced deoxyribonucleic acid degradation in spheroplast preparations.
Spheroplasts are insensitive to colicin E(2) and do not show deoxyribonucleic acid (DNA) degradation even in the presence of massive amounts of E(2). However, when both endonuclease I and E(2) were present, spheroplast DNA was degraded by an endonucleolytic activity which gave rise primarily to double-strand DNA cleavages, producing fragments having an average molecular weight of 9 x 10(6). Pan...
متن کاملObservations on the Fine Structure of Spheroplasts of Rhodospirillum Rubrum
Spheroplasts of the photosynthetic bacterium Rhodospirillum rubrum were prepared from cultures grown in either the presence or absence of light. Cells were converted into spheroplasts by using lysozyme and Versene and fixed in a sucrose-veronal-acetate buffer mixture containing osmium tetroxide. Some preparations were shadow-cast and examined whole; others were embedded in Epon 812 and sectione...
